Dry Packing (Alternative Method):

Add the dry adsorbent directly into the column.

Gently tap the column to ensure even packing.

Carefully add solvent along the column walls to avoid disturbing the packed adsorbent.

 

Condition the Column:

Once packed, pre-elute the column with solvent to remove air pockets and equilibrate the adsorbent.

 

Load the Sample:

Dissolve the sample in a small amount of the mobile phase.

Gently apply the sample on top of the packed adsorbent using a pipette or syringe.

Allow the sample to adsorb into the stationary phase.

 

Add the Mobile Phase:

Carefully add solvent to the top of the column without disturbing the adsorbent bed.

Open the stopcock to begin elution, maintaining a steady flow rate.

 

Collect Fractions:

Collect the eluted solvent in sequential fractions using test tubes or vials.

Monitor the fractions using UV light, TLC (thin-layer chromatography), or other analytical methods.

 

Optimize the Solvent System:

Start with a single solvent or a low-polarity solvent.

Gradually increase the polarity if compounds do not elute.
